The 21-year-old midfielder was stretchered off in the 24th minute of the 1-0 defeat having gone down injured during the move which led to game’s only goal.
The injury occurred when Charlton’s tricky winger Lloyd Sam ran directly at O’Toole, twisting one way and then the other in quick succession. O’Toole appeared to turn too sharply when trying to track his man and was left in a crumpled heap as Sam crossed for Nicky Forster to head home.
““We’ve lost John-Joe O’Toole to a serious knee injury,” said U’s boss Aidy Boothroyd.
“He has put all his weight on his left knee and has said he felt something crack. That’s not want you want to be hearing.”
