Read, who was the club's reserve team manager, took over from former Ipswich Town midfielder Mick Stockwell after he quit in November 2007.
Andy Lawrence, who was Read's assistant, has been put in charge while Woodbridge advertise for a new manager.
The Woodpeckers, who are second from bottom of the Ridgeons League Premier Division, are away to Histon Reserves on Saturday.
