With teams who finished 13th in League One or above seeded, as well as those from the north and south kept apart, the 410-mile round trip to the Welsh border is one of the furthest the U’s could have got.
John Trewick’s side finished 16th in League Two last season, having finished rock-bottom of League One the previous campaign.
Colchester also drew Hereford away in the FA Cup second round last season, the U’s scraping a 1-0 win at Edgar Street courtesy of John-Joe O’Toole’s last gasp winner.
Colchester were knocked out of the Carling Cup at the first hurdle last season with a 2-1 home defeat by Leyton Orient.
Elsewhere, Ipswich Town face an even longer trip to Exeter, while newly-promoted Norwich City will host Gillingham.
